Düzce İli Migren Tanısı Almış Hastalarda ENOS Ekzon (G894T) Polimorfizminin Araştırılması

Özet
Amaç: Düzce ili migrenli hastalarda eNOS ekzon (G894T) polimorfizm-hastalık ilişkisinin belirlenmesi amaçlanmıştır. Gereç ve Yöntem: Çalışmaya 51 migrenli hasta ve 51 sağlıklı birey dâhil edilmiştir. Bireylerin DNA'sı izole edilerek, eNOS ekzon (G894T) polimorfizmini içeren gen bölgesi PCR yöntemi ile çoğaltıldıktan sonra, Mbo I restriksiyon enzim kesim reaksiyonu ile genotip analizleri gerçekleştirilmiştir. Bulgular: Migren ve kontrol grupları arasında yaş (p0,70), cinsiyet (p0,529), homosistein (p0,250) ve trigliserit (p0,575) değerleri açısından anlamlı bir farklılık bulunmamıştır. Kontrol ve migren grubu arasında eNOS ekzon (G894T) polimorfizmi açısından anlamlı bir farklılık (p0,0001) gözlenirken, migren auralı (MA) ve migren aurasız (MO) grubu arasında anlamlı bir farklılık (p0,377) saptanmamıştır. Bu polimorfizm ile hem cinsiyet (p0,760) hem de aile öyküsü arasında (p0,380) bir farklılık belirlenmemiştir. Sonuç: Düzce ili popülasyonundaki migrenli hastalar ile eNOS ekzon (G894T) polimorfizmi arasında anlamlı bir ilişki (p0,0001) belirlenmiştir. Gelecek araştırmalarda, migren oluşumunu tetikleyici diğer aday genlerin de (MTHFR, ACE vb) yöremizdeki daha fazla hasta serisinde çalışılması öngörülmektedir.
Objective: Determination of relation between disease and polymorphism has been aimed through investigation of eNOS exon (G894T) polymorphism frequency in Duzce province. Materials and Methods: In this study, 51 patients with migraine and 51 healthy individuals were included. The genomic DNA of individuals were isolated and the gene region including eNOS exon (G894T) polymorphism were amplified by PCR (Polymerase Chain Reaction) method and analysed after digesting by MboI restriction enzyme. Results: There was no difference between migraine and control groups among age (p0,70), sex (p0,529) homocysteine (p0,250) and triglyceride (p0,575) values. While the eNOS exon (G894T) gene polymorphism between migraine and control groups were significant (p0,0001), no difference was observed between MA and MO groups for related gene polymorphism (p0,377). Also, no difference was detected between this polymorphism and both sex (p0,760) and family history(p0,380). Conclusion: A significant relationship was determinated between eNOS exon (G894T) gene polymorphism and patients with migraine (p0,0001). Research on other candidate genes (MTHFR, ACE etc.), having role as trigger in migraine, is being planned to be studied using a great number of patients.
