Testis üzerine cisplatinin apoptotik etkisine karşı timokinonun koruyucu etkisi

Özet
Bu tez çalışmasıyla, biyolojik etkileri olduğu bilinen ve son yıllarda yapılan araştırmalarda önemi artan timokinon ile bir antitümör ilaç olan Cisplatinin testis dokusu üzerindeki olumsuz etkilerini ortadan kaldırmayı veya en aza indirmeyi amaçladık. Bu amaçla 29 adet, Wistar albino rat rastgele 1. Kontrol (n = 4), 2. Sham (n = 4), 3. Cisplatin (n = 7), 4. Cisplatin + Timokinon (n = 7), 5. Timokinon (n = 7), gruplarına ayrıldı. 2. grup Sham grubuna 10 gün boyunca gavaj yoluyla mısır yağı (500 ?l) verildi. 3. Cisplatin grubuna 1.5 mg / kg / gün Cisplatin 3, 5 ve 7. günlerde intraperitoneal enjeksiyonla uygulandı. 4. Timokinon + Cisplatin grubuna; gavaj yolu ile mısır yağı (500 ?l) + timokinon 50 mg / kg / gün ve cisplatin 3, 5 ve 7. günlerde 1.5 mg / kg / gün intraperitoneal olarak uygulandı. 5.grup Timokinon grubuna 10 gün boyunca gavaj yoluyla timokinon (50 mg / kg / gün + mısır yağı (500 ?l)) verildi. Deneyin ilk aşamasında ratların vücut ağırlıkları ölçüldü. Deneyin 10. günde anestezi altında deneklerin testisleri alındı, histopatolojik incelemeler için alınan testisler rutin işlemlere tabi tutuldu. Epididimisten alınan örneklerden sperm sayımları yapıldı. Spermler hemotoksilen eozin ile boyandı. Testisler HE, Gomori trikrom ve PAS boyaları ile boyandı. Cleaved Caspase - 3 antikoru ile immunohistokimyasal boyama yapıldı. Işık mikroskobik inceleme sonucunda; timokinonun cisplatinin neden olduğu testiküler hasarı ve germ hücre apoptozisini azalttığı tespit edildi.
With this thesis, we aimed to eliminate or minimize the adverse effects of Cisplatin, an antitumor drug, Cisplatin, on the testicular tissue with thymoquinone, which is known to have biological effects and has increased in recent years. Corn oil (500 ?l ) was administered to the second group of Sham group by gavage for 10 days. 3. Cisplatin group 1.5 mg / kg / day cisplatin 3, 5 and 7. days were administered by intraperitoneal injection. 4. Thymoquinone + Cisplatin group; corn oil (500 µl) by gavaj + thymoquinone 50 mg / kg / day and cisplatin 3, 5 and 7. on days, 1.5 mg / kg / day was administered intraperitoneally. The 5th group of Timokinon group was given thymocinone 50 mg / kg / day + corn oil (500 ?l) by gavage for 10 days. In the first stage of the experiment, the body weights of the rats were measured. On the 10th day of the experiment, testes of the subjects were taken under anesthesia, and the testicles taken for histopathological examinations were routinely processed. Sperm counts were performed from samples taken from the epididymis. Sperms were stained with hematoxylin eosin. The testicles were stained with HE, Gomori trichrome and PAS paints. Immuno histochemical staining was performed with Cleaved Caspase - 3 antibody. As a result of light microscopic examination; It was found that thymoquinone reduced testicular damage and germ cell apoptosis caused by cisplatin.
