Özet
Astım, Dünya Sağlık Örgütü verilerine göre 339 milyon insanda görülen kronik bir solunum sistemi hastalığıdır. Prevalansındaki bu artış, astımın halk sağlığı ve sağlık giderleri açısından önemli bir yük oluşturduğunu göstermektedir. Bu bağlamda, geleneksel ve tamamlayıcı tıp uygulamalarından biri olarak arı ve arı ürünlerinin sağlık destekleyici ve tedavi edici amaçla kullanımını kapsayan apiterapi uygulamalarının astım hastalığının semptomları üzerinde etkilerinin araştırılması önem kazanmaktadır. Apiterapi kapsamında kullanılan apiair (kovan havası) uygulaması, arı kovanı içerisindeki havada bulunan uçucu bileşikler sayesinde solunum yolu üzerinde potansiyal destekleyici etkiler sunabileceği öne sürülen bir yöntemdir. Bu tez çalışmasında, ovalbumin ile indüklenen deneysel astım modeli üzerinde apiair ve kestane balı uygulamalarının, astım kaynaklı olarak akciğer dokusu üzerinde oluşabilecek fibrotik değişiklikler (fibrozis), oksidatif stres parametreleri, boy uzaması, vücut ağırlığı ve vücut kitle indeksi ile genel sağlık durumu üzerine etkileri incelenmiştir. Ayrıca arı temelli terapilerin potansiyel risklerinden olan alerji ve anafilaksi olasılığı değerlendirilmiştir. Bu amaçla tasarlanan deney modeli 8 gruptan oluşturuldu (Kontrol, astım, apiair, kestanebalı, astım+kestanebalı, astım+apiair, apiair+kestanebalı, astım+apiair+kestanebalı). Astım modeli ovalbumin ile indüklendi. Gruplara tedavi amacı ile periyodik olarak 30 gün boyunca oral gavaj ile kestane balı ve inhalasyon yöntemi ile apiair uygulaması sonrası deney sonlandırıldı. Ratların akciğer dokuları gomori trikrom yöntemi boyanarak ışık mikroskobunda fibröz doku birikimi yönünden incelendi, oksidatif stres parametreleri (TAS, TOS, OSİ) ve IgE ile boy uzaması, kilo alımı ve vücut kitle indeksi izlemi yapılarak genel sağlık bulguları değerlendirildi. Sonuçlar en yüksek fibröz doku artışının astım grubunda olduğunu göstermiştir (p <0 .001). En düşük fibröz doku skoru ise apiair grubunda tespit edilmiştir. Kestane balı uygulanan gruplar orta düzey fibröz doku skoru sergilemiştir. Gruplar arasında oksidatif stres belirteçleri ile IgE düzeylerinde anlamlı fark izlenmemiştir. Bu bulgular apiterapi temelli uygulamaların astım ve akciğer fibrozisi gibi kronik solunum yolu hastalıklarında akciğer dokusu hasarını sınırlama potansiyeli taşıdığını desteklemektedir.
Asthma is a chronic respiratory disease affecting 339 million people worldwide according to World Health Organization data. The increasing prevalence of asthma demonstrates its significant burden on public health and healthcare expenditures. In this context, research into the effects of apitherapy applications defined as the use of bees and bee products for health-supporting and therapeutic purposes on asthma symptoms has become increasingly important. Within apitherapy, the use of "apiair" (beehive air), which involves inhalation of volatile compounds naturally present in hive air, has been proposed as a method that may offer potential supportive effects on the respiratory system. In this thesis, the effects of apiair and chestnut honey applications on fibrotic changes (fibrosis) in lung tissue, oxidative stress parameters, body length, body weight, body mass index, and general health status were investigated in an ovalbumin-induced experimental asthma model. Additionally, potential risks associated with bee-based therapies, such as allergy and anaphylaxis, were evaluated. The experimental design consisted of 8 groups (Control, Asthma, Apiair, Chestnut Honey, Asthma + Chestnut Honey, Asthma + Apiair, Apiair + Chestnut Honey, Asthma + Apiair + Chestnut Honey). Asthma was induced using ovalbumin. For therapeutic purposes, chestnut honey was administered via oral gavage and apiair was delivered via inhalation periodically for 30 days. At the end of the trial, the rats' lung tissues were stained using the Gomori trichrome method and examined under a light microscope for fibrous tissue accumulation. Oxidative stress parameters (TAS, TOS, OSI) and IgE levels were measured, and assessments of body length, weight gain, and body mass index were conducted to evaluate general health findings. The results demonstrated that the highest increase in fibrous tissue occurred in the asthma group (p <0 .001). The lowest fibrosis score was observed in the apiair group. Groups treated with chestnut honey exhibited moderate fibrosis scores. No significant differences were observed among the groups with respect to oxidative stress markers or IgE levels. These findings support the potential of apitherapy-based applications to limit lung tissue damage in chronic respiratory diseases such as asthma and pulmonary fibrosis.
