ÖZET Düzce Yöresinde Yayılış Gösteren Lamiaceae Familyası Üyesi Üç Türün (Prunella vulgaris , Thymus longicaulis subsp. longicaulis ve Teucrium chamaedrys subsp. chamaedrys) Antimikrobiyal Aktiviteleri Üzerine Araştırmalar Kaan ÇETİN Düzce Üniversitesi Lisansüstü Eğitim Enstitüsü, Biyoloji Anabilim Dalı Yüksek LisansTezi Danışman: Prof.Dr.Başaran DÜLGER Temmuz, 2022 100 sayfa Bu çalışmada antimikrobiyal aktiviteyi tespit etmek amacıyla, Düzce ilinden toplanan Prunella vulgaris L., Thymus longicaulis subsp. longicaulis C.Presl., Teucrium chamaedrys subsp. chamaedrys L. (Lamiaceae) bitkilerinden elde edilen etanol, etil asetat ve kloroform ekstraktları, Pseudomonas aeruginosa, Escherichia coli, Salmonella typhimurium, Streptococcus pyogenes, Proteus vulgaris, Klebsiella pneumoniae, Staphylococcus aureus, Enterococcus faecalis, Bacillus cereus, Bacillus subtilis bakteri kültürleri ve Candida albicans, Candida tropicalis, Kluyveromyces marxianus, Cryptococcus neoformans ve Debaryomyces hansenii, maya kültürlerine karşı antimikrobiyal aktiviteleri disk difüzyon yöntemiyle araştırılmıştır. Prunella vulgaris L. bitkisinin en yüksek antibakteriyel aktivitesi etanol ekstratında P. aeruginosa (14 mm) bakterisine karşı ölçülmüştür. Bitkinin en yüksek antifungal aktivitesi ise etanol ekstraktında K. marxianus (15mm) maya kültürüne karşı saptanmıştır. Thymus longicaulis subsp. longicaulis C.Presl., bitkisinin en yüksek antibakteriyel aktivitesi etanol ekstratında B. subtilis (15 mm) bakterisine karşı ölçülürken, en yüksek antifungal aktivitesi ise yine etanol ekstraktında C. neoformans (12 mm) maya kültürüne karşı ölçülmüştür. Teucrium chamaedrys subsp. chamaedrys L. bitkisinin en yüksek antibakteriyel aktivitesi etanol ekstratında S. aureus (14 mm) bakterisine karşı bulunmuştur. En yüksek antifungal aktivitesi ise yine etanol ekstraktında K. marxianus (17 mm) maya kültürüne karşı ölçülmüştür. Ek olarak üç bitki türünün etanol ekstraklarının kimyasal karakterizasyonu Gaz Kromatografisi Kütle Spektroskopisi (GC/MS) tekniği ile araştırılıp değerlendirilmiştir. (Lamiaceae) collected from Düzce province were investigated for their antimicrobial activities against the bacterial cultures; Pseudomonas aeruginosa, Escherichia coli, Salmonella typhimurium, Streptococcus pyogenes, Proteus vulgaris, Klebsiella pneumoniae, Staphylococcus aureus, Enterococcus faecalis, Bacillus cereus, Bacillus subtilis and the yeast cultures; Candida albicans, Candida tropicalis, Kluyveromyces marxianus, Cryptococcus neoformans, Debaryomyces hansenii by disc-diffusion method. The highest antibacterial activity of Prunella vulgaris L. was measured against P. aeruginosa (14 mm) bacterium in the ethanol extract. Besides, the highest antifungal activity of the plant was determined in the ethanol extract against the yeast culture as K. marxianus (15 mm). While the highest antibacterial activity of Thymus longicaulis subsp. longicaulis C.Presl. plant was measured against B. subtilis (15 mm) bacterium in the ethanol extract, the highest antifungal activity was found against the yeast culture C. neoformans (12 mm) in the ethanol extract. Likewise, the highest antibacterial activity of Teucrium chamaedrys subsp. chamaedrys L. plant was detected against S. aureus (14 mm) bacterium while the highest antifungal effect was determined in K. marxianus (17 mm) as the yeast cultures in the ethanol extracts of the plant. In addition to all these, chemical characterization of the ethanol extracts of three plant species used in this study was investigated and evaluated by Gas Chromatography Mass Spectroscopy (GC/MS) technique.
