Öğe A case with pseudohypoaldosteronism type 1 and investigation of molecular genetic etiology(Duzce University Medical School, 2017) Bolu, Semih; Doğan, Mustafa; Eröz, Recep; Yüce, Hüseyin; Mermerci, Asuman; Özmerdivenli, RecepPseudohypoaldosteronism is a salt-wasting pattern that manifests with hyponatremia, hyperkalemia, and metabolic acidosis, and is the result of aldosterone peripheral nonresponse in renal tubule cells. Peripheral resistance development may occur as a result of mutations in the mineralocorticoid receptor or epithelial sodium channel; it can also develop as a secondary to infection, uropathy and receptor resistance due to drug use. Type 1 PHA is inherited as both autosomal dominant (sporadic-renal form) and autosomal recessive (systemic form). Systemic pseudohypoaldosteronism type 1 is autosomal recessive and the most severe form. Loss of function in one of the three subunits of the epithelial sodium channel (EnaC) is responsible for the disease (the alpha subunit (SCNN1A; 12p13), the beta subunit (SCNN1B; 16p12.2-p12.1), and the Gamma subunit (SCNN1G; 16p12). We present a patient who was diagnosed the primary pseudohypoaldosteronism type 1 for contribution to the literature, which is a rare disease and can be confused with other diseases caused by salt loss. © 2017, Duzce University Medical School. The aim of this study is to investigate the effects of exogenous galanin given with exercise on anxiety. Material and Methods: In this study 96 adult male Wistar Albino rats were used. Rats were divided into 8 groups with twelve animals, and 4 of the groups were given regular exercise for 6 weeks. Galanin was administered intraperitoneally (IP) and intracerebroventricular (ICV) in half of the groups and saline was injected into the control group of each group with the same methods. At the end of the sixth week, open field and elevated plus maze tests were applied to the rats, and these tests recorded with the camera. The recordings of the both tests were analyzed, and total distance and speed values, the time spent in the open field test and the time spent in the closed arm of elevated plus maze were compared. Results: In the open field test were determined that in general, the total distance (p<0.001) and the speed increased (p<0.001) in rats injected with ICV compared to the rats injected with IP. There is no significant difference was detected in terms of exercise and galanin interaction. Also the total distance (p=0,032) and the speed increased (p=0,010) in rats injected with ICV compared to the rats injected with IP. In elevated plus maze test, exercise and galanin interaction were not found to be statistically significant, however the interaction between the injection pattern and the given substance was found to be significant in terms of time spent in the closed arm (p=0.020) and speed (p=0.032). Conclusion: Although it has been shown that central administration of galanin may have anxiolytic activity, this effect has not changed with exercise, and also it has been determined that exercise with galanin has no effect on this activity. © 2018, Duzce University Medical School. All rights reserved.Öğe Galanin ve egzersizin ratlarda uzaysal hafıza performansına etkisi(Düzce Üniversitesi, 2016) Mermerci, Asuman; Özmerdivenli, Recep; Gümüştekin, KenanÖğrenme, edinilen deneyimlerin kalıcı hale gelmesi ve davranışa yansımasıdır. Bellek ise tüm bu öğrenilenlerin depolanması işlemidir. Öğrenme; beynin birçok bölümünün işbirliği içinde gerçekleştirdiği bir işlem olup, birçok farklı yöntemle edinilebilir. Bellek kayıtları da beynin farklı kısımlarınca depo edilebilir. Bu çalışmanın amacı; egzersizle birlikte verilen eksojen galaninin hafıza ve öğrenme üzerine etkilerinin gösterilmesi olup, hafıza ve öğrenmeyi azaltıcı yönde etki yapan galaninin etkilerinin egzersizle azaltılabilir olup olmadığının araştırılmasıdır. Çalışmamızda 108 adet Wistor Albino cinsi rat kullanıldı. Ratlar 12' şerli 9 gruba ayrıldı ve 5 gruba 6 hafta boyunca düzenli egzersiz yaptırıldı. Grupların yarısına ıntraperitoneal yarısına da ıntracelebroventriküler şekilde galanin ve her grubun kendi kontrol grubuna da salin enjekte edildi. 6. Hafta sonunda morris su tankında 4 gün süreli eğitime alınan ratlara, 5. gün hafıza ve öğrenme testi uygulanarak, kamera ile kayıtları yapıldı. Kayıtlar bilgisayar programına aktarılarak istatistik analizleri yapıldı. Çalışma öncesi ve sonrası ratlardan alınan kanlar santrifüj edilerek eliza testi yapıldı. Kan sonuçları SPSS programında post - hoc testi uygulanarak istatistik analizi yapıldı. Çalışmamızın sonucunda elde ettiğimiz bulgulara göre, düzenli egzersiz yapan ratların kontrol grubuna göre platformu bulma süresinin kısalmış olduğu ve ICV galanin verilen ratlarda platformu bulma süresinin kontrol gruplarına göre artmış olduğu, düzenli egzersizin ratlarda hafıza ve öğrenme potansiyelini artırdığı bunun yanısıra ICV verilen galaninin hafıza ve öğrenme potansiyelini azalttığı gözlendi. IP verilen galaninin ise hafıza ve öğrenme üzerine herhangi bir etkisi tespit edilemedi. Düzenli egzesizin, galaninin hafıza ve öğrenme üzerine olan olumsuz etkisini azaltamadığı, yapılan eliza testi sonucuna göre egzersiz yapan ratlarda kanda galanin seviyesini arttığı belirlendi. Sonuçlarımıza dayanarak, düzenli egzersizin galaninin hafıza ve öğrenme üzerine olan olumsuz etksini azaltamadığını, egzersizin hafıza ve öğrenme üzerine olan olumlu etkisini ise galaninin etkilerini azaltarak ya da bloke ederek gerçekleştirmediğini söyleyebiliriz. Ayrıca çalışmamız sonuçlarından yola çıkarak, alzheimer gibi hafıza ve öğrenme bozukluğu yaşanan hastalıklarda, tedavi yaklaşımlarına destek olunabileceğini düşünmekteyiz.
