Öğe Analysis of HPV Genotypes and Liquid-Based Cervical Cytology: Results from a Tertiary Academic Center in Northwestern Turkey(Natl Inst Infectious Diseases, 2021) Caliskan, Emel; Coskun, Sinem Kantarcioglu; Ozturk, Cihadiye Elif; Cangur, Sengul; Onail, BinnurHigh-risk human papilloma virus (hrHPV) plays an important role in cervical cancer. The aim of this study was to investigate the distribution of HPV genotypes in the region and to correlate it with liquid-based-cytology (LBC) and colposcopic biopsy results. Furthermore, the potential relationship between HPV infections and bacterial vaginosis (BV) was investigated. HPV genotypes were determined using real-time PCR. LBC, biopsies, and BV examinations were performed by the Pathology and Cytology. Consecutive cervical specimens of 409 women who underwent both cytology and HPV-DNA tests were included in the study. A total of 172 (42.1%) patients were positive for HPV-DNA; of these, 107 (26.2%) had hrHPV. The most common HPV genotypes were HPV 59, 16, 33, 52, and 51, at 16.6%, 15.9%, 13.4%, 13.4%, and 8.9%, respectively. Epithelial cell abnormality was detected in 11.5% of LBC test results. The genotypes of HPV 33, 56, 66, and 68 were found at a higher rate in patients with epithelial cell abnormalities than in those with no detected abnormalities. Bacterial vaginosis was found in 24 patients (5.9%). HPV-DNA positivity was observed to be statistically higher in patients with BV than in those without BV.Öğe Clinicopathological Features of Extranodal Lymphomas: 15 Years' Experience of a Single Center(Duzce Univ, 2021) Coskun, Sinem Kantarcioglu; Nazlioglu, Hulya Ozturk; Buyukuysal, CagatayObjective: In this study, we aimed to evaluate the localization and histopathological diagnosis, and clinicopathologic characteristics of primary extranodal lymphomas. Methods: The pathology reports between 2001 and 2015 in the archives of Uludag University Faculty of Medicine Pathology Department were reviewed and all cases with an extranodal lymphoma diagnosis were analyzed. The information about the diagnosis, tumor localization, symptoms at presentation, presence of B symptoms, lymphocytosis and anemia, chronic infection and chronic disease and concomitant secondary malignancy, tumor diameter, the involvement of another extranodal organ, lymph node, bone marrow, spleen, liver, stage of the disease, serum B2 microglobulin, LDH, albumin levels, sedimentation rate were documented. The localization, histopathological types, age groups, male/female ratios in cases of primary extranodal lymphoma were evaluated. Results: Total sum of 1743 patients were diagnosed with lymphoma. 480 (%27,53) of these cases were extranodal lymphomas. The most commonly encountered locations of extranodal lymphomas were the skin and the gastrointestinal system. There were 226 primary extranodal skin, 90 gastrointestinal system, 44 central nervous system, 8 genitourinary system, 50 head and neck, 18 musculoskeletal system and soft tissue, 9 mediastinum, 3 bronchus, 10 orbital, 2 liver, 6 pancreas, 4 omentum, 8 endocrine system, and 2 breast located cases. 237 of the primary extranodal lymphomas from our work were mature T/NK celled neoplasias. There were 250 patients with mature B cell lymphoma, 5 cases of Hodgkin lymphoma, and 5 cases of precursor lymphoid neoplasia. Conclusions: The data from our series were coherent with the literature. Due to the small number of cases with some localization and some histopathological diagnosis, no significant results could be reached about these entities.Öğe Investigation of the effects of pentoxifylline and alpha tocopherol treatment on recovery in rats with Achilles tendon rupture(Wiley, 2024) Toker, Mustafa; Karaduman, Zekeriya Okan; Arican, Mehmet; Turhan, Yalcin; Coskun, Sinem Kantarcioglu; Dalaslan, Rasit Emin; Celik, MuecahitAlthough the Achilles tendon is the largest and strongest tendon in the body, healing of the Achilles tendon is the most common injury, and this process is difficult due to poor tendon circulation; moreover, the underlying mechanism has not been fully elucidated. In our study, we aimed to investigate the effects of pentoxifylline and alpha-tocopherol administered separately or in combination on rats with Achilles tendon injury. Forty-eight male Wistar rats weighing 230 +/- 30 g were used in the study. The rats were randomly divided into eight groups of six animals each. Tendons were evaluated histopathologically and biomechanically. According to the statistical analysis, the vascularity density in the pentoxifylline group on day 14 was significantly greater than that in the other groups (p < 0.05). The collagen arrangement in the pentoxifylline and alpha-tocopherol groups on day 14 was found to be firmer and smoother than that in the control group (p < 0.05). The collagen arrangement in the pentoxifylline group on day 28 was greater than that in the other groups (p < 0.05). The biomechanical results were significantly greater in all groups (p < 0.05). Pentoxifylline contributed to tendon healing both through neovascularization in the early period and by improving collagen orientation in the late period, while alpha-tocopherol had a positive effect on collagen orientation in the early period. No beneficial effects were observed when pentoxifylline and alpha-tocopherol were used together. We believe that further research is needed to understand the effects of this combination therapy on tendon healing.Öğe Investıgatıon of the effects of treatment with enoxaparın sodıum and hyperbarıc oxygen therapy on the recovery of rats wıth achılles tendon rupture(Bmc, 2024) Aytekin, Cafer Erman; Turhan, Yalcin; Karaduman, Zekeriya Okan; Arican, Mehmet; Saglam, Sonmez; Coskun, Sinem Kantarcioglu; Uludag, VeyselPurpose In this study, we aimed to investigate the effects of hyperbaric oxygen therapy and enoxaparin sodium, which are known to accelerate bone tissue healing as well as tendon and soft tissue healing, on the healing of Achilles tendon rupture. Methods Thirty-six rats were used in the present study. All rats were divided into groups of nine. The groups were the enoxaparin sodium group, enoxaparin sodium and hyperbaric oxygen group, hyperbaric oxygen group and control group. After 21 days, the process was completed, and the rats were sacrificed. Achilles tendon samples were evaluated histopathologically. Results The groups were compared according to the results of statistical analysis based on the histopathological data. There was no significant difference between the groups in terms of acute inflammation (p = 0.785) or chronic inflammation (p = 0.827) scores, but there were significant differences in neovascularization (p = 0.009), proliferation (p < 0.001) and fibrosis (p = 0.006) scores. Conclusion Our study showed that the use of enoxaparin sodium and hyperbaric oxygen had a positive effect on the healing of the Achilles tendon. Based on these results, we believe that the use of enoxaparin sodium and hyperbaric oxygen therapy after Achilles tendon rupture will be beneficial for healing and preventing complications.Öğe KRAS, BRAF, PIK3CA mutation frequency of radical prostatectomy samples and review of the literature(Taylor & Francis Ltd, 2021) Bahcivan, Atike; Gamsizkan, Mehmet; Coskun, Sinem Kantarcioglu; Cangur, Sengul; Yuksel, Alpaslan; Ceyhan, Aysegul; Onal, BinnurObjective The molecular basis of prostate cancer is highly heterogeneous. Our study aimed to perform the mutation analysis of KRAS, BRAF, PIK3CA, and immunohistochemical (IHC) evaluation of EGFR, HER2, p16, and PTEN to demonstrate new areas for targeted therapies. Methods A total of 24 prostatectomy samples diagnosed with adenocarcinoma were analyzed by microarray hybridization. Also, these samples were IHC stained for EGFR, HER2, P16, and PTEN. The cases were divided into two groups based on low and high Gleason scores. All findings were compared with the clinicopathological parameters of the patients. Results While KRAS mutation was in 3/24 (12.5%) of our cases, BRAF and PIK3CA mutations were not detected. There was no significant difference between the groups in terms of KRAS mutation frequency. HER2 was immunohistochemically negative in all samples. There was no correlation between EGFR, P16 immunopositivity, and clinicopathological features. Conclusion KRAS mutation frequency is similar to those in Asian populations. BRAF and PIK3CA mutation frequencies have been reported in the literature in the range of 0-15% and 0-10.4%, respectively, consistent with our study findings. HER2 immunoexpression is a controversial issue in the literature. EGFR and p16 expressions may not correlate with the stage.Öğe Radiologic and histopathologic effects of favipiravir and hydroxychloroquine on fracture healing in rats(Springer, 2024) Tekce, Giray; Arican, Mehmet; Karaduman, Zekeriya Okan; Turhan, Yalcin; Saglam, Sonmez; Yucel, Muecahid Osman; Coskun, Sinem KantarciogluFracture healing is a process in which many factors interact. In addition to many treatments, physical and biological therapy methods that affect different steps of this process, there are many biological and chemical agents that cause fracture union delay. Although the number of studies on fracture healing is increasing day by day, the mechanism of fracture healing, which is not fully understood, still attracts the attention of all researchers. In this study, we aimed to investigate the effects of favipiravir and hydroxychloroquine used in the treatment of COVID-19. In this study, 48 male Wistar rats weighing 300 +/- 50 g were used. Each group was divided into eight subgroups of six rats each to be sacrificed at the 2nd and 4th weeks and evaluated radiologically and histologically. Favipiravir (group 1), hydroxychloroquine (group 2), favipiravir + hydroxychloroquine (group 3), and random control (group 4) were used. A statistically significant difference was observed between the 15th day histological scoring averages of the groups (p < 0.05). Although there was no statistically significant difference between the 15th day radiological score distributions of the groups (p > 0.05), we obtained different results in terms of complete bone union distributions and radiological images of the fracture line. Although favipiravir has a negative effect on fracture union in the early period, favipiravir may have a positive effect on fracture union in the late period. We did not find any effect of hydroxychloroquine on fracture union.
