Öğe Bazı yerel entomopatojen fungusların Amerikan beyaz kelebeğine Hyphantria cunea (Drury) (Lepidoptera: Arctiidae) karşı laboratuvar koşullarındaki insektisidal aktivitesi(2020) Karabörklü, Salih; Altın, Nedim; Yıldırım, Ismet; Öztemiz, Sevcan; Sadıç, Esat; Aydın, ÖmerAmerikan beyaz kelebeği, Hyphantria cunea meyve bahçeleri, ormanlık alanlar, park ve bahçelerde önemli zararlara neden olmaktadır. Bu çalışma Beauveria bassiana ve Metarhizium anisopliae türü entomopatojen fungusların H. cunea üzerindeki insektisidal aktivitelerinin belirlenmesi amacıyla laboratuvarda gerçekleştirilmiştir. Beauveria bassiana ve M. anisopliae türü toplam 10 izolat 3, 4 ve 5. dönem 10’ar adet H. cunea larvalarına 1x105 konidi/ml konsantrasyonda püskürtme yoluyla uygulanmıştır. Deneme 4 tekrarlı yürütülmüş olup en yüksek insektisidal aktivite 7. gün sonunda 3. dönem larvalarda belirlenmiştir. Beauveria bassiana YK23 izolatı 3. dönem larvalarda %89.72 oranında, M. anisopliae YK45 izolatı ise %76.39 oranında öldürücü etki göstermiştir. Diğer izolatlar da ise %55 ila %73.89 arasında değişen oranlarda öldürücü etki belirlenmiştir. Beauveria bassiana YK23 ve M. anisopliae YK43 izolatları 4. dönem larvalarda sırasıyla %77.78 ve % 72.22 oranında öldürücü etki göstermiştir. Aynı izolatlar 5. dönem larvalarda sırasıyla %60 ve %42.5 oranında ölüme neden olmuştur. Elde edilen sonuçlar zararlı ile mücadelede erken larva dönemlerinin seçilmesinin daha uygun olacağını göstermektedir.Öğe BIOCONTROL POTENTIAL OF Beauveria bassiana AND Metarhizium anisopliae ISOLATES FROM TURKEY AGAINST Hyphantria cunea (DRURY) (LEPIDOPTERA: ARCTIIDAE) LARVAE UNDER LABORATORY AND FIELD CONDITIONS(Univ Federal Uberlandia, 2022) Karabörklü, SalihHyphantria cunea is one of the most important pest insects causing significant damage in many plant species. The present study aimed to evaluate the insecticidal activity of Turkey isolates of Beauveria bassiana and Metarhizium anisopliae against H. cunea larvae under laboratory and field conditions. B. bassiana isolates YK16, YK23 and YK26, and M. anisopliae isolates YK41 and YK45 were sprayed onto the larvae of H. cunea at the respective doses of 1x10(5) and 1x10(6) conidia mL(-1) and monitored for seven days. Fungal isolates, bioinsecticide Bacillus thuringiensis and chemical insecticide diflubenzuron were used in field studies. Trials were carried out in five replications. All B. bassiana isolates caused 100% mortality on first instar larvae in laboratory trials. Mortality ratios ranged from 100 to 96% on second instar larvae. M. anisopliae isolates YK45 and YK41 caused 88 and 84%, and 81.33 and 77.11% mortalities for the first and second instar larvae, respectively. The mortality rates fluctuated between 91.78-72.89% for B. bassiana on third instar larvae in laboratory conditions. However, M. anisopliae isolates YK45 and YK41 caused 77.11 and 60.22% mortality on third instar larvae, respectively. In the field trials, B. bassiana YK23 displayed promising insecticidal activity with 80.60% mortality on second instar larvae of H. cunea. Other isolates as well caused mortalities ranging from 60.77 to 49.55%. The results revealed that some isolates of B. bassiana and M. anisopliae have potential to control H. cunea larvae. However, additional detailed studies need to be carried out to increase their effectiveness in field conditions.Öğe THE CHEMICAL COMPOSITION OF CYCLOTRICHIUM ORIGANIFOLIUM ESSENTIAL OIL AND ITS INSECTICIDAL ACTIVITY AGAINST FOUR STORED-PRODUCT INSECT PESTS(Publ House Bulgarian Acad Sci, 2019) Karabörklü, Salih; Azizoğlu, Uğur; Yılmaz, Semih; Ayvaz, Abdurrahman; Akdeniz, MehmetStored-product insect pests, Acanthoscelides obtectus, Tribolium castaneum, Ephestia kuehniella and Plodia interpunctella cause serious problems all over the world and cause significant losses in agricultural crops every year. In the present study, the essential oil from Cyclotrichium origanifolium (La bill.) Manden. & Scheng. (Labiatae) was extracted, chemically characterized and tested on these insect pests. Essential oil yield was estimated to be 0.95% (v/w) with major components being pulegone (74.60%) and menthone (16.61%). Limonene (1.12%) and isopulegone (1.09%) followed the major components. Significant insecticidal activity was achieved by treating the insect pests with the essential oil. Complete mortality was obtained on A. obtectus and T. castaneum adults at the 40 mu l L-1 air concentration during 24 h exposure period. More pronounced effect was observed on the adults of E. kuehniella and P. interpunctella. Nearly complete mortality was obtained at the 8 mu l L-1 air concentration. The results indicated that C. origanifolium essential oil displayed promising effects in controlling stored-product insect pests.Öğe Cloning and expression of cry2Aa from native Bacillus thuringiensis strain SY49-1 and its insecticidal activity against Culex pipiens (Diptera: Culicidae)(Academic Press Ltd- Elsevier Science Ltd, 2017) Yılmaz, Semih; Azizoğlu, Uğur; Ayvaz, Abdurrahman; Temizgül, Rıdvan; Atcıyurt, Zehra Büşra; Karabörklü, SalihBacillus thuringiensis (Berliner) (Bt) is well known for having toxicity against pest insects because of their ability to form endospores and broad-range activity of their parasporal inclusions. In this study, a new member of cry2A gene from previously characterized native B. thuringiensis SY49-1 strain was cloned, expressed and used for its activity against Culex pipiens (Diptera: Culicidae) larvae. The sequence analysis of the cloned cry2A gene revealed that it encodes a polypeptide of 633 as residues with 99% identity to Cry2Aa protein with expected molecular weight of 70.7 kDa. Bacillus thuringiensis delta-endotoxin nomenclature committee designed our sequence as Cry2Aa18 being a new member of Bt toxins. Bioassays against last instar larvae of C pipiens indicated that Cry2Aa18 has considerable toxicity with LC50 of 630 jig ml(-1). In order to prevent the spread of infectious diseases mediated by C. pipiens, this newly characterized cry2Aa18 gene could constitute as an important biological control tool for controlling mosquito larvae living in freshwater systems and can be used as a good alternative for minimizing the use of chemicals. All rights reserved.Öğe Düzce İli Fındık Depolarında Görülen Zararlı Böcekler ve Patojen Fungusların Tanımlanması(2018) Karabörklü, Salih; Altın, NedimZararlı böcekler ve hastalık etmenleri fındığın depolanması sırasında önemli ekonomik kayıplara nedenolmaktadır. Bu çalışma fındığın depolanması sırasında görülen zararlıların ve hastalık etmenlerinin belirlenmesiamacıyla yapılmıştır. Bu amaçla, Düzce il merkezi ve ilçelerinden 49 depodan fındık örnekleri alınmış velaboratuvarda incelenmiştir. İncelenen örneklerin %51.02’sinde (25 örnekte) böceklerden kaynaklı zarar tespitedilmiştir. Bu örneklerde Curculio nucum, Plodia interpunctella ve Rhyzopertha dominica zararı tespitedilmiştir. Böcek yoğunluğu ise 0-6.67 (%) arasında değişiklik göstermiştir. İncelenen örneklerin % 97.96’sında(48 örnekte) ise fungal hastalık etmenlerine rastlanmıştır. Fungal patojenlerden kaynaklı hastalık şiddeti(çürüklük oranı) ise 5.56-20.67 (%) arasında değişiklik göstermiştir. İç çürüklüğe neden olan fungal etmenler,Aspergillus sp., Penicillium sp., Alternaria sp., Fusarium sp., Chaetomium sp., ve Trichothecium sp. olarakbelirlenmiştir. Bu çalışma neticesinde fındıkta zarar oluşturan ve çürüklüğe neden olan önemli zararlı ve fungaletmenler belirlenmiştir. For this purpose, Bt kurstaki HD1 was incubated at 30 degrees C in T3 medium at 200 rpm for seven days. Lyophilized spore-crystal mixture (5000 mu g mL(-1)) was mixed with 50% honey solution and supplied to 0-24 h old T. evanescens adults as a nutrient to ensure the ingestion of the toxins by the parasitoids. The results indicated that spore-crystal mixture of Bacillus thuringiensis subsp. kurstaki (Btk) HD1 did not induce considerable decrease in parasitization performance and longevity of T. evanescens adults. Thus, it can be concluded that Btk HD1 products can safely be used together with egg parasitoid T. evanescens in integrated pest management system to compensate the deficiency of each control tactic alone.Öğe Expression of cry1Ab gene from a novel Bacillus thuringiensis strain SY49-1 active on pest insects(Soc Brasileira Microbiologia, 2016) Azizoğlu, Uğur; Ayvaz, Abdurrahman; Yılmaz, Semih; Karabörklü, Salih; Temizgül, RıdvanIn this study, the cry1Ab gene of previously characterized and Lepidoptera-, Diptera-, and Coleoptera-active Bacillus thuringiensis SY49-1 strain was cloned, expressed and individually tested on Ephestia kuehniella (Lepidoptera: Pyralidae) and Plodia interpunctella (Lepidoptera: Pyralidae) larvae. pET-cry1Ab plasmids were constructed by ligating the crylAb into pET28a (+) expression vector. Constructed plasmids were transferred to an Escherichia coli BL21 (DE3) strain rendered competent with CaCl2. Isopropyl beta-D-1-thiogalactopyranoside was used to induce the expression of cry1Ab in E. coli BL21(DE3), and consequently, similar to 130 kDa of Cry1Ab was obtained. Bioassay results indicated that recombinant Cry1Ab at a dose of 1000 mu g g(-1) caused 40% and 64% mortality on P. interpunctella and E. kuehniella larvae, respectively. However, the mortality rates of Bt SY49-1 strains' spore-crystal mixture at the same dose were observed to be 70% on P. interpunctella and 90% on E. kuehniella larvae. The results indicated that crylAb may be considered as a good candidate in transgenic crop production and as an alternative biocontrol agent in controlling stored product moths. In the current study, nematodes were isolated from soil samples and fungi associated with these free-living soil nematodes were determined. Large subunit (LSU) rDNAs of nematode-associated fungi were amplified and sequenced to construct phylogenetic trees. Nematode-associated fungi were observed in six nematode strains belonging to Acrobeloides, Steinernema and Cephalobus genera in different habitats. Malassezia and Cladosporium fungal strains indicated an association with Acrobeloides and Cephalobus nematodes, while Alternaria strains demonstrated an association with the Steinernema strain. Interactions between fungi and free-living nematodes in soil are discussed. We suggest that nematodes act as vectors for fungi.Öğe Fındık (Corylus avellana) Yaprakbitlerinin Düzce’deki Mevcut Durumunun Belirlenmesi(2020) Şen, Ahmet; Karabörklü, SalihBu çalışma fındık (Corylus avellana) yaprakbitleri,Myzocallis coryli Goeze ve Corylobium avellanae Schrank(Hemiptera: Aphididae)’nin Düzce’deki durumununbelirlenmesi amacıyla gerçekleştirilmiştir. Farklı yükseltiaralıklarına sahip 29 fındık bahçesinde Mayıs-Ekimperiyodu boyunca inceleme ve sayım yapılmıştır. M. corylive C. avellanae’nin yaygınlık oranları sırasıyla %100ve %96.55 olarak bulunmuştur. M. coryli’nin ocak vesürgündeki bulunma oranı Haziran-Eylül dönemi için %100,Mayıs ayı için %96.90 ve %94.77, ve Ekim ayı için %76.55ve %68.28 olarak hesaplanmıştır. Corylobium avellanae’deise ocak ve sürgündeki en yüksek bulunma oranları%38.97 ve %31.38 ile Haziran ayında belirlenmiştir. Diğeraylardaki bulunma oranları ise %25.86-31.38 ve %17.77-%27.82 arasında değişiklik göstermiştir. Myzocallis corylive C. avellanae’nin yoğunluklarının Haziran ayında zirveyeulaştığı, Temmuz ayından itibaren de kademeli bir şekildeazaldığı görülmüştür. Sürgün ve yaprak başına düşen enyüksek zararlı yoğunluğu Haziran ayında kaydedilmiş ve M.coryli için 49.22 ve 10.8 adet, C. avellanae için ise 8.92 ve1.96 adet olarak hesaplanmıştır. Yalnızca Eylül ayında artanrakıma bağlı olarak M. coryli’nin yoğunluğunda istatistiksel açıdan önemli bir azalma görülmüştür. Sürgünve yaprak başına düşen yoğunluk 0-249 m içinsırasıyla 30.57 ve 6.51 adet iken bu yoğunluk500 m ve üzerinde sırasıyla 13.65 ve 3.13adete kadar gerilemiştir. Her iki türündeyoğunluğunun ekonomik zarar eşiğinin altıdakaldığı belirlenmiştir.Öğe Mosquitocidal Potential of Native Bacillus thuringiensis Strain SY49-1 against Disease Vector, Culex pipiens (Diptera: Culicidae)(Malaysian Soc Parasitology Tropical Medicine, 2017) Azizoğlu, Uğur; Yılmaz, Semih; Ayvaz, Abdurrahman; Karabörklü, Salih; Atcıyurt, Zehra BüşraMosquitoes are regarded as troublesome organisms worldwide due to their parasitic and pathogenic aspect causing malaria, yellow fever, dengue, West Nile and filariasis. Bacillus thuringiensis (Bt) products have effectively been used for decades in controlling the mosquito mediated diseases and also decreasing the chemical input into the environment. In the present study, biocontrol potential of previously characterized Bt SY49-1 strain was investigated on Culex pipiens larvae. Fourth instars of C. pipiens were subjected to spore/delta-endotoxins (50, 100, 250 and 500 mu g mL(-1)) of Bt SY49-1 strain, carrying cry genes active against Lepidoptera, Diptera, Coleoptera and Nematodes. The spore/delta-endotoxin mixture caused 100% mortality on the larvae at the dose of 500 mu g mL(-1). PCR results indicated that Bt SY49-1 harbors Lepidopter-Dipter specific cry2A gene as well as Lepidopter specific cry1Ab, cry1Aa/Ad, cry1C, cry9A, cry9C, Lepidopter-Dipter-Coleopter specific cry1B, and Nematode specific cry5 gene. Results indicated the potential usage of Bt SY49-1 in preventing the breeding of C. pipiens and the spread of diseases resulting therefrom.Öğe Native Entomopathogenic Fungi Isolated from Duzce, Turkey and their Virulence on the Mealworm Beetle [Tenebrio molitor L. (Coleoptera: Tenebrionidae)](Univ Philippines Los Banos, 2019) Karabörklü, Salih; Altın, Nedim; Keskin, YeşimNative entomopathogenic fungi were isolated from Duzce province of Turkey and their virulence was tested against Tenebrio molitor larvae using direct spraying and grain spraying methods. Forty-five isolates were obtained. Beauveria bassiana was obtained from 24.07% of the soil samples and Metarhizium anisopliae from 11.11% of the soil samples. The level of insecticidal activity of the isolates fluctuated between 20% and 100%, when directly sprayed on the larvae. In the direct spray application, the most effective isolates were B. bassiana YK11, YK16, M. anisopliae YK43, YK44 and YK45. These isolates caused 100% mortality on the larvae of T. molitor at 168 h. In 12 isolates, there was 100% mortality at 240 h under laboratory condition. Lethal time (LT50) of B. bassiana and M. anisopliae isolates varied from 10.48 to 127.40 h. Virulence of the fungal isolates in 3% (w/v) aqueous suspension was lower than that in the 10% aqueous (w/v) suspension in the grain spray application. Virulence of fungal isolates was significant in 10% aqueous suspension and 93.33% and 100% mortality were observed in B. bassiana YK23 and YK26, respectively, at 240 h. B. bassiana and M. anisopliae isolates showed promising effectiveness for the control of insect pests.Öğe Native entomopathogenic nematodes isolated from Turkey and their effectiveness on pine processionary moth, Thaumetopoea wilkinsoni Tams(Taylor & Francis Ltd, 2015) Karabörklü, Salih; Ayvaz, Abdurrahman; Yılmaz, Semih; Azizoğlu, Uğur; Akbulut, MikailTwo native entomopathogenic nematodes were isolated from soil samples in Eastern Mediterranean region of Turkey and characterized based on 28S rDNA region. BLASTN homology and phylogenetic analysis of SK17 and SK-71 isolates indicated 98% and 99% identity to Steinernema affine and Steinernema feltiae, respectively. The results were constructed by neighbour-joining and bootstrap tree methods. Efficacy of S. affine (SK-17 strain) and S. feltiae (SK-71 strain) was tested against the larvae of pine processionary moth, Thaumetopoea wilkinsoni Tams, and remarkable mortality rates were obtained. Both strains caused complete mortality upon application of 500 IJs in foliar tests. However, the same strains caused 30% and 33% mortality at 80 IJs/cm(2) in soil applications. It was concluded that these native strains could be considered as potential biocontrol agents for reducing the damage caused by T. wilkinsoni larvae.Öğe PHYLOGENETIC RELATIONSHIPS OF INSECT-ASSOCIATED FREE-LIVING RHABDITID NEMATODES FROM EASTERN MEDITERRANEAN REGION OF TURKEY(Corvinus Univ Budapest, 2016) Azizoğlu, Uğur; Karabörklü, Salih; Ayvaz, Abdurrahman; Yılmaz, SemihFree-living nematodes play an important role in nutrient cycling. The study was designed to characterize insect-associated free-living soil nematodes in Rhabditida order from Eastern Mediterranean region of Turkey using their 28S rDNA sequences and phylogenetic analysis. A total of 105 soil samples were taken from different habitats. Galleria baiting technique was used to obtain free-living nematodes. The nematodes were harvested from cadavers of last instar Galleria mellonella larvaeafter trapping process. Free-living soil nematodes were detected in 14 soil samples (13.3%). PCR products obtained from 28S rDNA of isolates were sequenced and phylogenetic trees created. BLASTN homology searches and phylogenetic tree results constructed by Neighbour Joining and Bootstrap Tree methods. Results showed that these insect-associated isolates were determined as species belonging to Rhabditidae (Rhabditis terricola and Mesorhabditis sp.), Chambersiellidae (Macrolaimus sp.) and Cephalobidae (Cephalobus sp., Pseudacrobeles sp. and Acrobeloides sp.) families in Rhabditida. Free-living nematodes interact with many arthropods and other invertebrate species. These interactions are also important for their use in biological control program. In this study, these interactions between free-living nematode-insect,-arthropod and-invertebrate are discussed.Öğe The potential of Beauveria bassiana and Metarhizium anisopliae in controlling the root-knot nematode Meloidogyne incognita in tomato and cucumber(Korean Soc Applied Entomology, 2022) Karabörklü, Salih; Aydınlı, Volkan; Dura, OnurMeloidogyne incognita is one of the most important plant parasitic nematodes. This study was conducted to determine the nematicidal potential of Beauveria bassiana and Metarhizium anisopliae against M. incognita. B. bassiana and M. anisopliae spore suspensions and bio -nematicide, Purpureocillium lilacinum were applied. B. bassiana and M. anisopliae revealed considerable nematicidal activity against M. incognita in tomato and cucumber. The gall index decreased gradually from 8.0 for control to 3.2, 2.0 and 2.2 for B. bassiana, M. anisopliae and P. lilacinum in tomato, respectively. The highest reduction (%) in gall formation (control index) was calculated as 75.2 % in M. anisopliae treated group for tomato. The gall index was 7.6 for control, but decreased to 3.6, 2.0 and 2.2 for B. bassiana, M. anisopliae and P. lilacinum in cucumber, respectively. The highest control index was 71.7 % for M. anisopliae in cucumber. The number of the second instar juveniles of M. incognita was recorded as 2240 for control. However, this value reduced to 508, 332 and 328 by B. bassiana, M. anisopliae and P. lilacinum in tomato, respectively. The highest control indexes for the second instar juveniles were 85.2 % and 85.3 % for M. anisopliae and P. lilacinum in tomato, respectively. Alike, the highest control indexes were 84.9 % and 85.7 % for the same applications in cucumber, respectively. B. bassiana and M. anisopliae displayed also positive effect on the number of leaves, dry and fresh root weights of tomato. The results showed that B. bassiana and M. anisopliae can be considered as an alternative.Öğe Recombinant entomopathogenic agents: a review of biotechnological approaches to pest insect control(Springer, 2018) Karabörklü, Salih; Azizoğlu, Uğur; Azizoğlu, Zehra BüşraAlthough the use of chemical pesticides has decreased in recent years, it is still a common method of pest control. However, chemical use leads to challenging problems. The harm caused by these chemicals and the length of time that they will remain in the environment is of great concern to the future and safety of humans. Therefore, developing new pest control agents that are safer and environmentally compatible, as well as assuring their widespread use is important. Entomopathogenic agents are microorganisms that play an important role in the biological control of pest insects and are eco-friendly alternatives to chemical control. They consist of viruses (non-cellular organisms), bacteria (prokaryotic organisms), fungi and protists (eukaryotic organisms), and nematodes (multicellular organisms). Genetic modification (recombinant technology) provides potential new methods for developing entomopathogens to manage pests. In this review, we focus on the important roles of recombinant entomopathogens in terms of pest insect control, placing them into perspective with other views to discuss, examine and evaluate the use of entomopathogenic agents in biological control.Öğe Yerel Beauveria bassiana ve Metarhizium anisopliae İzolatlarının Myzocallis coryli ve Corylobium avellanae Üzerindeki Etkinliği(2020) Karabörklü, Salih; Altın, NedimFındık yaprakbiti, Myzocallis coryli ve fındık yeşil afidi, Corylobium avellanae fındıkta zararoluşturan böcekler arasında yer almaktadır. Bu çalışma, Beauveria bassiana ve Metarhiziumanisopliae türü entomopatojen fungusların M. coryli ve C. avellanae üzerindeki öldürücü etkilerininbelirlenmesi amacıyla yürütülmüştür. Laboratuvar çalışmalarında nimf ve erginlere 1x105 konidi ml1 doz püskürtülmüş ve 7 gün boyunca denemeler takip edilmiştir. Uygulama süresi sonunda B.bassiana YK26, YK16, YK11 ve M. anisopliae YK45 izolatları sırasıyla M. coryli üzerinde %81.09,%73.24, 58.25 ve %52.45 oranlarında öldürücü etki göstermiştir. EPF izolatlarının C. avellanaeüzerinde oldukça etkili oldukları görülmüş ve en etkili izolatlar %88.9, %96.75 ve %98.08 öldürmeoranları ile B. bassiana YK26, YK11 ve M. anisopliae YK38 olmuştur. Arazi çalışmalarında ise B.bassiana ve M. anisopliae izolatları 1x106 konidi ml-1 konsantrasyonda M. coryli’ye uygulanmış ve enyüksek öldürme oranı %48.37 ile B. bassiana YK26 izolatında görülmüştür. Bu izolatı %44.69 ve%38.49 öldürme oranları ile B. bassiana YK16 ve M. anisopliae YK45 izolatları takip etmiştir. Yaprakbitleriyle mücadelede B. bassiana ve M. anisopliae izolatlarının kullanılabileceğine yönelik bulgularelde edilmiştir.
