ICP-MS Analysis and Validation by Microwave Digestion System for Determination of Heavy Metals in Allergy and Cancer Drugs Taken Orally
(Duzce University, 2023) Köse, Elif; Dalmaz, Aslıhan; Özak, Sezen Sivrikaya
The drug, one of the most important products that cannot be replaced in the field of health, prevents all threats to human health when used in place and dosage. Therefore, it has an important place in public health. Determination of the amounts and impurities of various elements used in drug production is an important parameter. According to the ICH Q3D(R1) international compliance conference guide, methods by which limit concentrations can be determined for elemental impurities by applying inductively coupled plasma (ICP) analysis to drugs are specified. In this study, the sample preparation step in the analysis of 23 elements in cancer and allergy drugs in tablet form was optimized by microwave digestion, and method development and validation studies were carried out for these drugs with inductively coupled plasma-mass spectrometry (ICP-MS). When the analysis results were evaluated, it was seen that the data obtained were by the limit values specified in the ICH. Detection and quantification limits of the developed method, and relative standard deviation values were determined, and recovery studies were carried out by adding standards at 4 different concentrations to determine the method's precision. The correlation coefficients obtained for heavy metals in this study ranged from 0.9993 to 1.000, while the detection limit was found to be between 0.001 and 1.756 µg/L. Thus, the reliability and precision of the validation study developed has been determined and it has been shown that this method can be used in similar drug samples.

Investigation of Permeability of Thiocolchicoside Through Transdermal Drug Delivery System Using Franz Diffusion Cell
(Sakarya Üniversitesi, 2024) Karagüzel, Hale; Özak, Sezen Sivrikaya; Dalmaz, Aslıhan
Recently, drug release applications through the skin have become very popular. One of the most remarkable of these drug release applications is transdermal drug release systems, which are drug release methods that allow the active substance to pass into the systemic circulation through the skin or artificial membranes. In this study, the optimization conditions required for the release and permeation tests of a gel drug containing the active substance thiocolchicoside were comparatively investigated using synthetic membranes without human or animal skin. For this purpose, the permeability of the gel drug in gel form and the active ingredient thiocolchicoside was carried out using Franz Diffusion Cell. As a result of the investigations, it was observed that the best synthetic membrane for the permeability of thiocolchicoside in the Franz Diffusion Cell was the Supor membrane. In addition, the method's relative standard deviation values, detection, and quantification limits were determined, and permeation studies were carried out. In this study, the correlation coefficient was found to be 0.9992, and the limits of detection and quantification were 0.026 and 0.078 µg/L. In this way, the sensitivity and reliability of the validation study were determined.

Use of ion chromatography method on the determination of some anions in the water collected from Sakarya, Turkey
(2019) Altundağ, Hüseyin; Ağar, Şerife; Altıntığ, Esra; Ateş, Asude; Özak, Sezen Sivrikaya
Abstract: In this study simultaneous analysis of seven different anions (Fluoride, Chloride, Bromide, Nitrite,Nitrate, Phosphate, and Sulfate) in13 different water samples collected from Sakarya/Turkey and it wasconducted with ion chromatography method. Analyzes were performed simultaneously using the ionchromatography method. Some validation tests and the optimum conditions for the determination of anions werestudied. The analysis of anions was accomplished by the dilution of the sample injection device. Samples wereused to adjust the terms of the device and the results were recorded. Ion chromatography (IC) is now consideredas an excellent technique for the analysis of ions in many samples.

A Vortex-Assisted Microextraction Based On Deep Eutectic Solvents for Determination of Four Parabens from Cosmetic Baby Oils and Optimization by Box-Behnken Design
    (2020) Özak, Sezen Sivrikaya
This study presents microextraction and determination of four parabens by vortex-assisted microextraction basedon deep eutectic solvents (DESs) in cosmetic baby oils. The most suitable DES, ChCl-ethylene glycol moleratios of DES were determined as 1:2 for use in this study. The effect of DES types has been explored attraditional one factor experiment at a time. The effect of DES volume, dilution solvent volume and vortex timeon extraction recovery were examined and optimized by the Box-Behnken design. After optimum conditionswere determined, vortex-assisted microextraction based on DES, 0.1-100 µg mL-1concentration in the range ofthe calibration curve was plotted for all parabens. The recovery values of parabens in cosmetic baby oil sampleswere in the range of 82.02-101.95% and the RSDs were varied from 1.64% to 2.91%. This developed method isvery suitable and effective method for the extraction of parabens from cosmetic products of similar structure.
